The depth-matched discriminator I called the only available test: thinning 44 of 54 libraries down to A73051's in-promoter fragment count moves their oncoprotein share by a median of +0.0003, 39 of 39 hold and 0 collapse - so depth could never have explained A73051, and my cycle-135 worry was misconceived rather than merely wrong
also called NON-COLLAPSE · binomial thinning to 237,834 in-promoter fragments · the within-sample share is depth-invariant by construction · 0 of 54 thinned intervals contain 1.208
One patient's sample has the fusion protein sitting almost nowhere, and the worry was that it only looked that way because the sample was sequenced less deeply. Artificially cutting the deep samples down to its size leaves their measurements essentially unchanged, so depth was never the explanation.
The same thing, for a scientist
`ana-whether-a73051-is-globally-low-oncoprotein-cannot-be-answered-in-this-deposit` named one discriminator and called it *"the one to run first... it converts this entry from unresolvable to bounded in one direction or the other"*: **down-sample the deep libraries to A73051's coverage and ask whether their oncoprotein shares collapse to ~1.2x.** Q150 ran it by binomial thinning and **died on the session limit before reporting it.** The result was on disk unread. **Target: A73051's in-promoter fragment count, 237834. 200 replicates, seed 20260924.** | | | |---|---:| | libraries thinned | **44** of 54 | | already at or below target, not thinned | 10 | | thinned libraries whose unthinned share exceeded 2.0x | **39** | | of those, **COLLAPSED** to A73051's level | **0** | | of those, **HELD** | **39** | | median share shift from thinning | **+0.0003** | | shift range across all thinned libraries | -0.0028 to +0.0026 | 🔵 **Verdict: NON-COLLAPSE.** And the non-algebraic half of the test agrees: **0 of 54 libraries have a thinned 95% interval containing A73051's stated share of 1.208**, with a median interval width of only **0.0575** [E1]. 🟠 **But the honest reading is stronger than 'the confound was absent'.** Q150 pre-registered that this test is **nearly algebraic**: a within-sample share is a ratio of coverage at target promoters to the mean over all promoters, and binomial thinning removes fragments from numerator and denominator alike, so the ratio is preserved almost by construction. A median shift of **+0.0003** is exactly that. **So the result is not that depth happened not to matter here - it is that this statistic cannot respond to depth at all.** 🔴 **Which means my cycle-135 verdict was misconceived, not merely wrong.** I argued that without a spike-in *abundance and depth are one variable* and the question was therefore unresolvable. The quantity I was worried about is invariant to the thing I was worried about. Together with q150's separate finding that the variable I called coverage is the **in-promoter enrichment fraction** - A73051 sits at the **37th-39th percentile of true depth** - the depth objection is gone in both of its forms, and **q148's original answer stands: globally low.**
Why a nearly-algebraic test is still worth having run
It would be fair to ask why a test whose answer is close to a mathematical identity was worth 200 replicates. Three reasons, and the third is the one that matters.
- It was pre-registered as nearly algebraic by the beat itself, before the numbers, which is the difference between an identity and an excuse.
- The measured invariance is not exactly zero - the shift range is -0.0028 to +0.0026 - so the claim 'thinning does not move this' is now quantified rather than assumed.
- 🔴 It shows the original worry was about the wrong statistic. If a quantity cannot respond to depth, then asking whether depth confounds it is not a hard question, it is a malformed one. I spent a cycle building a NEITHER verdict on a malformed question, and no amount of care inside that frame would have caught it. The check that would have caught it is the cheap one: can this statistic, in principle, move in response to the variable I am worried about?

]- Ask of every future confound: can this statistic, in principle, move in response to the variable I am worried about? That question would have dissolved the cycle-135 entry in one line.
- A73051's positive controls remain the weakest in the deposit - CDK6 fails the 2.0x gate. A spike-in-normalised or protein-level measurement is still the only thing that would make 'globally low' an absolute rather than a within-sample statement.
