HB11;19 (CVCL_8227) is KMT2A::MLLT1, not KMT2A::AFF1 — and it is the most menin-dependent line in the panel

also called HB(11;19) · HB11.19 · HB 11;19 · HB1119 · CVCL_8227 · ACH-001736

HB11;19 is often listed with the infant leukaemia cell lines, but its fusion partner is a different gene from the one this project studies. It is also the line most dependent on menin, which complicates the story.

The same thing, for a scientist

HB11;19 (CVCL_8227, DepMap ACH-001736) is a B-ALL line with **KMT2A::MLLT1 (MLL-ENL)**, not KMT2A::AFF1: its name encodes t(11;19), and Cellosaurus annotates the fusion from the founding paper that identified the trithorax homolog involved in 11q23 translocations and from an array-CGH study of fusion-positive lines. Its age and sex are both recorded as **unspecified**, so it cannot be classified as infant-derived or not. It carries CDKN2A p.Arg80Ter homozygous and two TP53 mutations (p.Thr125Met, p.Arg248Trp). It is one of the eleven lines in this project's DepMap KMT2A-rearranged panel, and in the MEN1 screen query run for this beat it returned the **strongest** gene effect of the five lines checked, -1.983.

The correction, stated plainly

Any inventory that lists HB11;19 among KMT2A::AFF1 material is wrong. The partner is MLLT1 1. This entry exists because the line's name (HB11;19) looks like the t(4;11) line names (RS4;11, MV4;11, AN4;11) and because it sits in the same screened panel, which is exactly the shape that produces a silent mis-assignment.

It also cannot be counted as infant material: Cellosaurus records SX Sex unspecified and AG Age unspecified 1. So of the eleven lines in the screened DepMap panel, ten have a recorded age and none of those ten is under one year, while the eleventh has no age at all. The weak-spot phrasing 'eleven cell lines, none infant-derived' should therefore read 'ten of eleven have a recorded age and none is infant; the eleventh is unknown'.

Why its menin dependency matters to the index-fusion argument

In the same API call that returned MEN1 gene effects for SEM (-0.959), SEM-K2 (-1.667), RS4;11 (-0.463) and MV4-11 (-1.198), HB11;19 returned -1.983 2. The most menin-dependent line in that set carries a partner that is not the index fusion, and the least menin-dependent carries the index fusion.

That is not evidence against the menin interface — the interface is on the KMT2A side and is shared by all these fusions, which predicts exactly this kind of partner-independence. It is evidence against using cross-line depth of MEN1 dependency as a partner-specific argument, which this graph has been close to doing.

How this could bite us back
  • Treating HB11;19 as a KMT2A::AFF1 line would import MLLT1 biology into index-fusion conclusions, the same error class the provenance beat measured at 54% of load-bearing experiments.
  • Two TP53 mutations and homozygous CDKN2A loss make this line a poor model for a disease with a near-silent genome, whichever partner it carries.
Still open
  1. How old was the HB11;19 donor? The founding report may state it; the database does not, and the answer decides whether the screened panel contains an infant line at all.
  2. Is the partner-independence of MEN1 dependency real across a larger set of KMT2A fusions? Five lines cannot answer it, and the answer bears directly on whether menin inhibitors should be expected to work equally across partners.

Cellosaurus accession CVCL_8227 records HB11;19 as a human cancer cell line of unspecified sex and unspecified age, with NCIt disease term 'B acute lymphoblastic leukemia with t(v;11q23.3) MLL rearranged', carrying a KMT2A-MLLT1 (MLL-ENL) gene fusion, homozygous CDKN2A p.Arg80Ter and TP53 p.Thr125Met and p.Arg248Trp mutations, DepMap identifier ACH-001736, and membership of the Cancer Dependency Map project.

    What this evidence cannot show

Fetched by accession 2026-09-19 with the primary name read back. The fusion annotation is attributed to the 1992 founding paper (PMID 1423624) and to an array-CGH study (PMID 16523483). The absence of age and sex is a gap in the record, not evidence that the donor was an adult.

Evidence that argues against this (1)
    • HB11;19 is widely described in the literature as an infant ALL line and is used as one. If that description is correct, then the screened DepMap panel does contain an infant line — carrying MLLT1 rather than AFF1 — and the weak spot's phrasing is wrong for a second, independent reason. The database's silence cannot settle this either way, and the founding paper does not report the donor's age in its title or abstract. Tkachuk DC et al. 1992

    What this evidence cannot show

Five values, one API release, no intervals retrieved, no lineage adjustment, and two of the five lines (SEM, SEM-K2) are one patient. With n=4 patients this is a range rather than a comparison, and no partner-specific inference should be drawn from it in either direction.

Evidence that argues against this (1)
    • A raw Chronos value is not an effect size relative to other genes, and this project has already had to correct itself twice on reading DepMap numbers — once for the myeloid-enriched index arm, once for comparator-SD scaling. Comparing five raw values across lines with different screen quality, growth rate and library is the least defensible use of these data, and this entry uses them only to refuse an inference, not to support one.
