PARP1 - the target whose inhibitors this disease was specifically shown to resist, and which three later papers combine anyway
also called PARP1 · ADPRT · PPOL · ARTD1 · poly(ADP-ribose) polymerase 1
PARP1 senses small DNA nicks and calls in repair; blocking it is a famous way to kill cancers that cannot repair DNA accurately. KMT2A-rearranged leukaemia repairs DNA perfectly well, which is exactly why one careful study found it resistant to PARP inhibitors - and why the later reports of benefit are all combinations, not single agents.
The same thing, for a scientist
PARP1 (P09874, 1014 aa) is the principal poly(ADP-ribose) polymerase of the DNA damage response. It is the clearest worked example in this beat of the difference between a target that is druggable and a target that is indicated. **The load-bearing negative:** Esposito et al. showed that AML driven by MLL fusions with dominant transactivation ability is **DDR-proficient and insensitive to PARP inhibition**, in contrast to AML1-ETO and PML-RARA leukaemias which are extremely sensitive because of suppressed HR-gene expression; inhibiting the MLL target HOXA9, which activates HR-associated genes, impaired the DDR and sensitised MLL leukaemia to PARP inhibitors [E2]. **The supporting negative:** neither t(4;11) product changes PARP1 levels or HR/NHEJ efficiency in a controlled knock-in [E5]. **The combination results:** PARP1 is reported required for the maintenance of MLL-AF9 leukaemias with PARP1 inhibitors enhancing the effect of cytotoxic drugs [E3], and olaparib potentiates conventional chemotherapy and DNMT inhibitors in MLL leukaemia cells with a rise in cells carrying more than 10 gamma-H2AX foci [E4]. These are consistent if read carefully: the disease is HR-proficient, so PARP inhibitor **monotherapy** has no synthetic-lethal handle, and the combination results are potentiation of exogenous damage rather than exploitation of an intrinsic defect. An entry that cited only the combinations would read as a PARP-inhibitor recommendation in a disease with a published resistance mechanism.
Reading the three results together
| result | design | reading |
|---|---|---|
| MLL fusions DDR-proficient, PARPi-insensitive; HOXA9 inhibition sensitises 2 | mouse and human AML, genotype comparison across fusion classes | there is no intrinsic HR defect to exploit; sensitivity has to be manufactured |
| PARP1 required for maintenance of MLL-AF9 leukaemias; PARPi enhances cytotoxic drugs 3 | mouse MLL-AF9 | a maintenance requirement is not synthetic lethality; the therapeutic claim is combination |
| Olaparib potentiates chemotherapy and DNMT inhibitors; >10 gamma-H2AX foci 4 | mouse primary and human MLL-AF9 line, colony and growth assays | potentiation of exogenous damage, single agents had 'little impact' |
The common structure: every positive result is a combination and every combination supplies the DNA damage from outside. That is a coherent pharmacological position and it is not the position the phrase 'PARP inhibitors work in MLL leukaemia' conveys.
The one KMT2A-specific PARP1 biology that is not about inhibitors
miR-181a is reported to enhance drug sensitivity of MLL-rearranged AML by increasing PARP1 acetylation 6. That is a post-translational regulation claim rather than a target claim and it is recorded here at moderate confidence from its abstract; it is the only paper found that treats PARP1 as a regulated node in this disease rather than as a drug target.

| subnuclear localization | NUCLEOLUS (resting, ~40% of total PARP-1) PLUS NUCLEOPLASM AND CHROMATIN; DAMAGE MOVES IT OUT OF THE NUCLEOLUS AND ONTO BREAKS. A single resting-state answer would be wrong for this protein, which is why the record is split. CONSTITUTIVE: approximately 40% of PARP-1 is nucleolar in unstressed cells, the remainder nucleoplasmic and chromatin-bound including on undamaged chromatin [E_q89_parp1_loc1][E_q89_parp1_loc2]. INDUCED: nucleolar PARP-1 delocalises to the nucleoplasm on DNA damage; PARP1 accumulates at break sites; auto-PARylation then drives dissociation from chromatin [E_q89_parp1_loc2]. CAUTION ON UNIPROT'S NUCLEOLUS ROW: its sole attribution, PMID 9518481, reports colocalisation of PARP with DNA polymerase alpha-primase AT THE NUCLEAR PERIPHERY of HeLa cells by confocal microscopy and does not mention the nucleolus in any text this beat could reach - see E_q89_parp1_loc3. The nucleolar call is well supported by other work; its UniProt citation is not the support. |
| localization shown for fusion | NO - nobody has localised PARP1 in any KMT2A fusion, in any lineage. What exists in a fusion context is a LEVEL measurement in the index fusion (MLL-AF4 and AF4-MLL AAVS1 knock-ins, no change in PARP1 protein, this entry's E5) and a FUNCTIONAL negative in MLL-fusion AML generally (DDR-proficient, PARP-inhibitor-insensitive, E2 / E_q89_parp1_loc4). Given that this graph carries a menin-PARP experimental proposal, the absent measurement is specific and cheap: does PARP1's nucleolar fraction change under menin inhibition? |

(2) AWAY FROM chromatin: auto-poly-ADP-ribosylation promotes DISSOCIATION from chromatin, and PARP1 is extracted from chromatin by VCP/p97 following sumoylation and ubiquitination - so the same damage that recruits it then evicts it. (3) OUT OF the nucleolus: nucleolar PARP-1 delocalises to the nucleoplasm in cells exposed to DNA-damaging agents, and that delocalisation is separable from automodification and is itself what sensitises cells to damage-induced apoptosis [E_q89_parp1_loc2]. }- PARP inhibitors are approved agents and available, which makes it easy to propose them here. The specific, published finding in this disease is monotherapy resistance with a stated mechanism, and every positive result is a combination that supplies the DNA damage from outside.
- Adding a PARP inhibitor to cytotoxic chemotherapy in an infant increases the genotoxic load in a patient whose disease began with a DNA double-strand break and who has a full lifetime for a second malignancy to appear. PARP inhibitors carry a recognised MDS/AML signal in adults treated for other cancers; in this population that signal is the disease.
- Clinical PARP inhibitors are PARP1/PARP2 agents. Any member-level claim from a compound in this entry is naming and not measurement, exactly as for the TOP2 isoforms elsewhere in this beat.
- The sensitising manoeuvre in the published mechanism is HOXA9 inhibition - which is also the differentiation-block mechanism. A combination designed on that logic cannot attribute its effect to the DDR arm rather than to differentiation.
- Is KMT2A::AFF1 infant B-ALL PARP-inhibitor insensitive in the same way as the myeloid MLL fusions tested? The load-bearing negative was generated in AML and the index disease has never been tested.
- Do HOXA-low KMT2A-rearranged cases, which this graph holds as a distinct group, lack the HOXA9-driven HR proficiency and therefore behave like the PARPi-sensitive fusions? That is a biomarker-selected hypothesis nobody has stated.
- Does a menin inhibitor, which collapses the HOXA programme, phenocopy HOXA9 inhibition and sensitise to PARP inhibitors? The mechanism published in 2015 predicts it and no menin-inhibitor plus PARP-inhibitor combination exists.
- Does PARP1's resting nucleolar fraction differ in KMT2A-rearranged blasts? This graph holds a menin-PARP experimental proposal and a large nucleolar block of entries; the measurement is one immunofluorescence with a fibrillarin co-stain in a fusion-positive and a fusion-negative line.
- Does menin inhibition move PARP1? Nucleolar-to-nucleoplasmic delocalisation is what sensitises cells to damage-induced apoptosis, so if a menin inhibitor delocalised PARP1 it would predict a synergy that nobody has tested - and if it does not, that is a clean negative for the combination.
- PARP1's caspase-cleavage fragments have separate compartments, as KMT2A's taspase1 fragments do. Has anyone ever asked whether the two proteolytic two-fragment systems interact - for instance whether the irreversibly DNA-bound PARP1 N-terminal fragment accumulates at the KMT2A breakpoint cluster region during the apoptotic window in which fusions are thought to form?

A naming fact. gene_exact:PARP1 AND organism_id:9606 AND reviewed:true returned exactly one row with gene_primary PARP1. The alternative name ARTD1 is used in part of the literature and is NOT in this record's gene-synonym field, so a symbol-keyed search misses it - one of the 8 records returned by (MLL OR KMT2A) AND PARP1 uses ARTD1 in its title.
Evidence that argues against this (1)
- The paralogue is the standing hazard: clinical PARP inhibitors are not PARP1-selective. Olaparib, niraparib, rucaparib and talazoparib all inhibit PARP1 and PARP2, so every result in this entry that comes from a compound is a PARP1/PARP2 result. Q9UGN5 is 583 aa against P09874's 1014, which makes an accession swap detectable but does nothing about the pharmacological ambiguity. UniProt Consortium 2026

Leukaemia driven by MLL fusions with dominant transactivation ability is proficient in the DNA damage response and insensitive to PARP inhibition, while AML driven by repressive fusion oncoproteins (AML1-ETO, PML-RARalpha) is extremely sensitive owing to suppressed expression of key HR-associated genes; genetic or pharmacological inhibition of the MLL downstream target HOXA9 impairs the DDR and sensitises MLL leukaemia to PARP inhibitors, and HOXA9 overexpression confers PARP-inhibitor resistance to AML1-ETO and PML-RARalpha transformed cells.

- The same group subsequently reported that olaparib potentiates conventional chemotherapy and DNMT inhibitors in MLL leukaemia cells, with single-agent treatments having little impact. Evidence that argues against this (1)
- A maintenance requirement for PARP1 in a leukaemia is not evidence of a synthetic-lethal vulnerability and does not contradict the finding that the same disease class is insensitive to PARP inhibition as monotherapy. In primary mouse leukaemia cells and a human MLL-AF9 line, olaparib combined with DNMT inhibitors or chemotherapy significantly reduced colony formation or cell growth while single-agent treatments had little impact; the combination induced cell-cycle block and apoptosis and significantly increased the proportion of cells with more than 10 gamma-H2AX foci and double-strand breaks.

Evidence that argues against this (1)
- Gamma-H2AX focus counts are the mechanistic evidence, and gamma-H2AX is induced by replication stress and apoptotic fragmentation as well as by double-strand breaks. A DNMT inhibitor plus a PARP inhibitor perturbs replication directly, so the focus increase is a weaker inference about break burden than it reads. Kosik P et al. 2021

Two NLS motifs at 207-209 and 221-226. The note records that PARP1 also binds NORMAL, UNDAMAGED chromatin, that auto-poly-ADP-ribosylation promotes dissociation from chromatin, that VCP/p97 extracts it after sumoylation and ubiquitination, and that PRKDC phosphorylation drives nucleus-to-cytosol translocation.

A curated composite drawn from more than twenty papers across four decades and many cell systems, none haematopoietic in the rows examined here. The evidence codes say a curator saw experiments; rule 91 and E_q89_parp1_loc3 below show that for at least the nucleolus row the cited paper is not about the fact.
Evidence that argues against this (1)
- It is tempting to read twelve ECO:0000269 attributions for 'Chromosome' as overwhelming evidence, and the number is doing less work than it looks. Nine of those twelve are cited in the note specifically for localisation TO SITES OF DNA DAMAGE - an induced, sub-second, lesion-proximal recruitment - and only one (PubMed:15607977) is cited for binding normal undamaged chromatin. So a count that reads as 'twelve independent demonstrations that PARP1 lives on chromatin' is largely twelve demonstrations that PARP1 GOES to broken chromatin, which is a different claim and the one this entry is at pains to separate. The constitutive chromatin association rests on far fewer citations than the row's evidence count suggests, and a reader grading confidence by attribution count would get this backwards. APPROXIMATELY 40% OF PARP-1 IS NUCLEOLAR IN UNSTRESSED CELLS, and nucleolar PARP-1 delocalises to the nucleoplasm in cells exposed to DNA-damaging agents. Rancourt and Satoh separated delocalisation from automodification: unmodified PARP-1 was first delocalised from the nucleolus with camptothecin, then activated with MNNG. Relative to MNNG alone, prior delocalisation induced EXTENSIVE AUTOMODIFICATION of PARP-1; DNA repair activity and intracellular NAD+ consumption were UNAFFECTED; but apoptotic cell formation INCREASED, and that increase was suppressed by inhibiting PARP-1 activity. The authors propose nucleolar PARP-1 participates in nucleolus-mediated stress sensing.

Abstract read, full text not read; the cell system is not named in the abstract and is therefore not named in the claim (rule 73). The 40% figure is quoted from the authors' own framing of their earlier work rather than re-derived here, and no error bar or method is given for it in the abstract. Not haematopoietic, no KMT2A fusion, and camptothecin/MNNG are pharmacological rather than physiological stresses.
Evidence that argues against this (1)
- The entry's own framing - constitutive nucleolar pool, damage-induced exit - may have the causality inverted, and the paper cannot exclude it. Camptothecin is a topoisomerase I poison that generates replication-associated breaks AND is well known to disrupt nucleolar organisation directly; the nucleolus is famously the first structure to reorganise under almost any stress. So 'PARP-1 leaves the nucleolus in response to damage' and 'the nucleolus dissolves and PARP-1 goes with it' predict the same observation, and the second does not require PARP-1 to be a stress sensor at all. The finding that resists this reading is the functional one: prior delocalisation changed the CONSEQUENCE of later activation - more automodification, more apoptosis, with repair activity and NAD+ consumption unchanged - which is a specific, non-trivial result that bulk nucleolar disintegration does not obviously predict. The compartment claim and the sensing claim should therefore be graded separately, and this entry grades only the compartment. UniProt Consortium 2026

PMID 9518481 shows PARP is physically associated with the catalytic subunit of the DNA polymerase alpha-primase tetramer, 'an association confirmed by confocal microscopy, demonstrating that both enzymes are co-localized at the NUCLEAR PERIPHERY of HeLa cells'; the interaction requires the integrity of PARP's second zinc finger and is maximal in S and G2/M. The word 'nucleolus' appears nowhere in the abstract.
Functional association of poly(ADP-ribose) polymerase with DNA polymerase alpha-primase complex: a link between DNA strand break detection and DNA replication. · Dantzer F, Nasheuer HP, Vonesch JL, de Murcia G, Ménissier-de Murcia J · Nucleic Acids Res 1998What this evidence cannot show
Bounded by what is readable. The abstract was read in full. The Europe PMC fullTextXML route returned HTTP 500 for PMC147507 and the PMC HTML page returns 18,257 characters that contain ZERO occurrences of 'nucleol' and only one each of 'periphery' and 'confocal' - i.e. the 1998 article body is served as a scanned PDF and the text is not reachable, so a nucleolar observation could exist in the figures. The correct statement is therefore 'the accessible text of the cited paper reports nuclear periphery and not nucleolus', graded moderate for that reason. This does NOT challenge PARP1's nucleolar pool, which is independently established (E_q89_parp1_loc2).
Evidence that argues against this (1)
- The obvious defence is that the nucleolus abuts the nucleoplasm and a 1998 confocal image of 'nuclear periphery' might have meant the perinucleolar region, so the curator may have read the figures correctly and this beat, unable to see them, is second-guessing. That defence is plausible and cannot be refuted from outside the paper - which is why this item is moderate rather than strong. But the alternative reading is worse for the annotation, not better: 'nuclear periphery' in confocal usage overwhelmingly means the nuclear ENVELOPE/lamina region, which is close to the opposite of the nucleolus in both position and biology, and this graph maintains a separate localization-nuclear-envelope-proximity-kmt2a-amino-terminus entry on the strength of that distinction. If the curator did mean the envelope and filed it as nucleolus, a wrong compartment has been propagated; if the curator meant perinucleolar, the annotation is right and its wording is misleading. Either way a reader cannot tell from the record, and that is the defect. UniProt Consortium 2026

In MLL-FUSION-DRIVEN acute leukaemia, PARP1 is the target that does not work, and the reason is a cell-state property: AML driven by MLL fusions with dominant transactivation ability is DNA-damage-response-PROFICIENT and insensitive to PARP inhibition, in contrast to AML1-ETO and PML-RARA leukaemias, which are extremely sensitive because they suppress homologous-recombination gene expression. Inhibiting the MLL target HOXA9, which activates HR-associated genes, impaired the DDR and sensitised MLL leukaemia to PARP inhibitors.
Synthetic lethal targeting of oncogenic transcription factors in acute leukemia by PARP inhibitors. · Esposito MT, Zhao L, Fung TK, Rane JK, Wilson A, Martin N, Gil J, Leung AY, Ashworth A, So CW · Nat Med 2015What this evidence cannot show
The fusions studied are MLL fusions with dominant transactivation ability - the paper's own phrase - and the comparator leukaemias are AML1-ETO/RUNX1-RUNX1T1 and PML-RARA. This is myeloid and largely murine-model work; it is NOT a dedicated human KMT2A::AFF1 experiment, and this project's standing finding is that 0 of 24 sampled load-bearing experiments are. This item re-states the entry's existing E2 with re-fetched identifiers, and is added here because the cell-state field needs a citation of its own; the identifiers were taken from a fresh PubMed efetch rather than copied from the sibling block.
Evidence that argues against this (1)
- This entry's own later evidence argues against reading the negative as final, and the tension should be visible rather than resolved by preferring the first paper. PARP1 is reported REQUIRED for maintenance of MLL-AF9 leukaemias with inhibitors enhancing cytotoxic drugs (this entry's E3), and olaparib plus DNMT inhibitors or chemotherapy reduced colony formation in primary mouse leukaemia and a human MLL-AF9 line (E4). Those are not contradictions of Esposito if read carefully - they are COMBINATION results and a maintenance requirement, against a MONOTHERAPY sensitivity claim - but they do mean 'PARP inhibition does not work in MLL leukaemia' is too broad, and the entry's title already concedes this by saying three later papers combine anyway. The localisation dimension adds a specific reason the question may not be closed: nobody has asked whether PARP1's nucleolar fraction, or its chromatin residence time, differs in a KMT2A-fusion blast, and a DDR-proficient cell with a differently partitioned PARP1 could respond differently to trapping agents than to catalytic inhibitors. CARM1 interacts with PARP1 and promotes PARylation at replication forks, stimulating PARP1 by enhancing its DNA binding and acting jointly with HPF1 - and this is INDEPENDENT of CARM1's methyltransferase activity

PARP1 has a CONSTITUTIVE nucleolar pool of approximately 40% of total protein that delocalises to the nucleoplasm on DNA damage, and the delocalisation itself sensitises cells to damage-induced apoptosis (PMID 19144573) (Q89 2026-09-21)
PARP1 binds NORMAL undamaged chromatin constitutively, is recruited to breaks on damage, and is then evicted by auto-PARylation and by VCP/p97 - three movements in two directions, so no single resting-state record is correct (Q89 2026-09-21)
